Overall survival in patients with pancreatic cancer receiving matched therapies following molecular profiling: A retrospective analysis of the Know Your Tumor registry trial

Pishvaian MJ, Blais EM, Brody JR, et al. - By performing this retrospective study of the Know Your Tumor (KYT) programme, researchers inquired if a longer median overall survival could be experienced by patients with pancreatic cancer whose tumours harboured actionable molecular alterations (defined as molecular changes for which there is clinical or strong preclinical evidence of a predictive benefit from a specific therapy) and who were treated with molecularly matched therapy vs similar patients who were not managed with molecularly matched therapy. The KYT programme involves US patients suffering from pancreatic cancer and allows patients to undergo commercially available multi-omic profiling to afford molecularly tailored treatment choices and clinical trial recommendations. A median follow-up of 383 days revealed significantly longer median overall survival of patients with actionable molecular alterations who received a matched therapy vs those who only received unmatched therapies. As per these real-world findings, survival in patients with pancreatic cancer can be substantially influenced by the adoption of precision medicine, and further prospective assessment is justified for molecularly guided therapies targeting oncogenic drivers and the DNA damage response and repair pathway.
