Identification of long non-coding RNA and circular RNA expression profiles in atrial fibrillation

Wu N, Li J, Chen X, et al. - Researchers explored long non-coding RNA (lncRNA) and circular RNA (circRNA) expression profiles in atrial fibrillation (AF) patients and for this purpose, they obtained atrial tissues from 7 patients with AF and 7 matched controls. By means of Hiseq/Proton RNA sequencing, they discovered lncRNA and circRNA expression profiles of atrial tissues. Using 35 pairs of AF patients and controls, they carried out validation by reverse transcription quantitative real-time PCR. With fold change > 1.5, significant differential expressions of 557 lncRNAs and 280 circRNAs were seen. Validation showed significantly different expression levels of an lncRNA Voltage Dependent Anion Channel 2 Pseudogene 2 (VDAC2P2) and two circRNAs chr13_41887361_41865736_-21625 and chr13_100368574_100301460_-67114. The implication of some pathways such as ribosome and chromatin modification, Rap1 signalling and cardiac muscle contraction, in the pathogenesis of AF was revealed in Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analysis. In this study, lncRNA and circRNA expression, as well as their interaction with mRNA and miRNA in AF, were described.
