HIV influences clustering and intracellular replication of hepatitis C virus

Goyal A, Perelson AS, Kandathil AJ, et al. - In view of a common encounter with HCV and HIV coinfection and the reports of an increased HCV viraemia and accelerated disease progression because of HIV, researchers here examined liver biopsies from monoinfected (n = 4) and HCV/HIV‐coinfected (n = 5) participants for HCV RNA, based on single‐cell laser‐capture microdissection. They assayed HCV RNA in 3,200 hepatocytes with information of spatial position. Although monoinfected and coinfected participants showed comparable median plasma HCV RNA level and the fraction of infected cells, the median HCV RNA per infected hepatocyte in monoinfected was significantly lower than in coinfected participants. A mathematical model of infection indicated that HIV facilitates higher intracellular accumulation of HCV RNA by hindering the export of HCV RNA. Per these findings, HIV coinfection affects intracellular accumulation of HCV RNA and the clustering of HCV‐infected cells, but to a less extent the fraction of HCV‐infected cells.
