Efficacy of live oral rotavirus vaccines by duration of follow-up: A meta-regression of randomized controlled trials

Clark A, et al. - Given a substantial body of evidence on the effectiveness of live oral rotavirus vaccines in different settings, researchers sought to synthesize the data to obtain robust estimates of effectiveness by the duration of follow-up in this meta-regression study. Findings suggested that instantaneous vaccine effectiveness (iVE) pooled for infant schedules of Rotarix and RotaTeq was 98% (95% credibility interval 93–100) 2 weeks after the last vaccination dose and 94% (87–98) after 12 months in settings with low mortality (15 observations). In high-mortality settings vs low-mortality settings, lower rotavirus vaccine effectiveness was evident, and it waned more rapidly. However, high-mortality settings showed an earlier peak age of disease indicating the likelihood of still achieving substantial benefit with live oral rotavirus vaccines.
